Cyclodextrin Therapy Reverses Advanced Atherosclerosis Safely at Home

What is Cyclodextrin?


Cyclodextrin, specifically 2-Hydroxypropyl-β-cyclodextrin (HPβCD), is a fascinating sugar molecule that has been making waves in the medical community, especially concerning heart and vascular health. It’s been shown to help clear arterial plaque, dramatically reduce inflammation, and may even assist in detoxifying the body from microplastics. Cyclodextrins are naturally occurring in minute amounts in starchy plants like corn and potatoes, but are extracted and concentrated for medical and industrial purposes by treating starches with certain enzymes. Because of their unique ring-shaped molecular structure—with a fat-attracting interior and a water-attracting exterior—they can “trap” fat-soluble substances such as cholesterol and plastics.

The family of cyclodextrins are widely useful and have been used in many industries for some time now. In the food industry, they are used to stabilize and enhance flavors, for example to make chewing gum and candy maintain their flavor for longer periods, or to remove cholesterol in some foods like reduced cholesterol eggs. In pharmaceuticals, they help to increase solubility and permeability of drugs for better bioavailability. They are used to encapsulate fragrance molecules for sustained release in perfumes, and actives in skincare products to prevent degradation and to increase skin permeation. Beyond personal care, they’re even used to clean polluted water and soil, bind silver to fabrics for antimicrobial clothing, and more.


Cyclodextrins and Heart Disease


Heart disease typically involves the buildup of cholesterol-filled plaques in the arteries, leading to atherosclerosis—a condition where arteries become narrowed and hardened due to plaque accumulation, which can lead to high blood pressure,  heart attacks and strokes. Remarkably, HPβCD has shown promise in rapidly reversing plaque buildup. Research indicates that it can dissolve cholesterol crystals within arterial plaques, effectively reducing their size and promoting arterial health. This process not only helps in clearing existing plaques but also prevents the formation of new ones. ​


In a pivotal study published in Science Translational Medicine, researchers investigated the effects of 2-hydroxypropyl-β-cyclodextrin (HPβCD) on mice models of atherosclerosis. The findings revealed that HPβCD treatment not only reduced the size of atherosclerotic plaques but also decreased cholesterol crystal load within the plaques. Notably, these benefits were observed even when subjects continued on a cholesterol-rich diet.


Clinical Research and Human Trials


The potential of this novel treatment is underscored by the initiation of human clinical trials. Cyclarity Therapeutics has launched a Phase 1 clinical trial to evaluate the safety and effectiveness of their proprietary cyclodextrin therapy, marking a big step forward from lab research to real-world use.

How It’s Used and Where to Get It


Although HPβCD can be administered intravenously by a medical professional, the easiest current treatment protocol consists of a daily “micro-enema” application, similar to what one might do for hemorrhoid treatment. A small amount of gel containing the HPβCD is inserted into the rectum and retained for a minimum of 30 minutes, but preferably overnight. Oral administration is not effective as the cyclodextrins are broken down by the digestive system and never make it into the bloodstream. They are however, easily absorbed by rectal tissues and immediately enter the bloodstream where they start their work. The cholesterol and plaques are then broken down by the kidneys and excreted without complication.


While this breakthrough treatment is still under investigation for pharmaceutical approval, HPβCD is currently available without a prescription and doesn’t require a doctor for administration.
